Ed Broth is the author of Stories From A Moron Real Stories Rejected by Real Magazines , published in January, 2005 by St. Martin s Press. The book consists of numerous prank letters Broth mailed to magazines and their responses. An example is his submission of an essay titled I Love Dogs to I Love Cats magazine. The book shares many similarities with the books of Ted L. Nancy . Both are prank authors who submit joke letters to real organizations and then publish their responses. The covers of their books are also similar, showing the title and author s name written childishly. As well, the books of both Broth and Nancy include a foreword by comedian Jerry Seinfeld , who claims in both books to have met with the authors. Procedure Inoculate MR VP Glucose phosphate broth with pure culture of test organism. Incubate the broth at 35 sup 0 sup C for 48 72 hours. After incubation add 5 drops of Methyl Red directly into the broth, through the sides of the tube. br Interpretation The development of stable red color in the surface of the medium indicates sufficient acid production to lower the pH to 4.4 and constitute a positive test. Since other organism may produce lesser quantities of acid from the test substrate , an intermediate orange color between yellow and red may develop. This does not indicate positive test. br Controls br Positive and negative controls should be run after preparation of each lot of medium. br Positive control Escherichia coli br Negative control Klebsiella br br Voges Proskauer test br Principle br It is used to determine the ability of some organisms to produce a neutral end product, acetyl methyl carbinol acetoin from glucose fermentation. The production of acetoin, a neutral reacting end product produced by members such as Klebsiella, Enterobacter etc., is the chief end product of glucose metabolism and form less quantities of mixed acids. The IMViC tests are a group of individual tests used in microbiology lab testing to identify an organism in the Coliform bacteria coliform group . A coliform is a gram negative, aerobic or facultative anaerobic rod which produces gas from lactose within 48 hours. The presence of some coliforms indicate fecal contamination. Except for the lowercase i , which is added for ease of pronunciation, each of the letters in IMViC stands for one of these tests. I is for indole M is for methyl red V is for Voges Proskauer, and iC is for citrate. It contains tryptophan , which under the action of enzyme tryptophanase is converted to an Indole molecule, pyruvate and carbon dioxide. The indole is then extracted from the broth by means of xylene. To test the broth for indole production, Kovac s reagent is added. A positive result is indicated by a Pink Red layer forming on top of the liquid. Methyl Red test Voges Proskauer test These tests both use the same broth for bacterial growth. The broth is called MRVP broth. After growth, the broth is separated into two different tubes, one for the Methyl Red MR test and one for the Voges Proskauer VP test. The pH indicator Methyl Red is added to one tube and a red color appears at pH s lower than 4.2, and indicated positive test. The VP test uses alpha naphthol and potassium hydroxide to indicate a positive or negative test. Citrate Test This test uses Simmon s citrate agar to determine the ability of a microorganism to use citrate as its sole carbon source.
