It is based on a modification of the Wright s stain Wright Giemsa stain pioneered by Bernard Witlin in 1970. It has advantages over the older Wright Giemsa staining technique, as it reduces the 4 minute process into a simplified 15 second operation, and allows for selective increased eosinophilic or basophilic staining depending upon the time the smear is left in the staining solutions. Diff Quik is utilized on material which is air dried prior to alcohol fixation rather than immersed immediately ie. wet fixed . The primary use of Romanovsky type stains is for cytoplasmic detail, such as intracytoplasmic mucins, fat droplets and neurosecretory granules. Extracellular substances, such as free mucin, colloid, ground substance, etc, are also easily stained, and appear metachromatic. In some laboratories, a computer performs an initial evaluation, indicating slides that are sufficiently normal that no human reading is needed, or pointing out areas that may be of particular interest for later examination. In many laboratories, cytotechnologists perform the initial evaluation. The cytotechnologist performs the secondary evaluation and determines whether a specimen is normal or abnormal. Abnormal specimens are referred to a pathologist for final interpretation. Different countries have different certification requirements and standards for cytotechnologists. In the United States there are currently two routes for certification, after earning a baccalaureate degree, individuals attend an accredited program in cytotechnology for 1 year, or you can attend a cytotechnology program that awards a baccalaureate degree in cytotechnology.
