Corticotropin releasing hormone CRH plays multiple roles in vertebrate species. In mammals, it is the major hypothalamic releasing factor for pituitary adrenocorticotropin secretion, and is a neurotransmitter or neuromodulator at other sites in the central nervous system. In non mammalian vertebrates, CRH not only acts as a neurotransmitter and hypophysiotropin, it also acts as a potent thyrotropin releasing factor, allowing CRH to regulate both the adrenal and thyroid axes, especially in development. Corticotropin releasing factor bindingprotein CRH BP is thought to play an inhibitory role in which it binds CRH and other CRH like ligands and prevents the activation of CRH receptor s. There is however evidence that CRH BP may also exhibit diverse extra and intracellular roles in a cell specific fashion and at specific times in development ref name PUB00011280 cite journal author Seasholtz AF, Valverde RA, Denver RJ title Corticotropin releasing hormonebindingprotein biochemistry and function from fishes to mammals journal J. Because ABP binds to T and DHT, these hormones are made less lipophilic and become concentrated within the luminal fluid of the seminiferous tubules. The higher levels of these hormones enable spermatogenesis in the seminiferous tubules and sperm maturation in the epididymis . ABP has the same amino acid sequence as sex hormonebinding globulin SHBG the difference is the site of production and the addition of different sugar moieties. ABP contains 403 amino acids, resulting in a molecular weight of 44,533. Its gene is located on chromosome 17 . ABP s production is regulated under influence of Follicle stimulating hormone FSH on Sertoli cell, enhanced by insulin , retinol , and testosterone . ABP may also be secreted in other organs for instance, mice produce a saliva ry ABP. Maltose BindingProtein MBP is a part of the maltose maltodextrin system of Escherichia coli , which is responsible for the uptake and efficient catabolism of maltodextrins. It is a complex regulatory and transport system involving many protein s and protein complexes. MBP has an approximate molecular mass of 42.5 kilodaltons. Use MBP is used to increase the solubility of recombinant protein s expressed in E. coli . In these systems, the protein of interest is often expressed as a MBP fusion protein , preventing aggregation of the protein of interest. The mechanism by which MBP increases solubility is not well understood. In addition, MBP can itself be used as an affinity tag for purification of recombinant proteins. The fusion protein binds to amylose columns while all other proteins flow through. The MBP protein fusion can be purified by eluting the column with maltose. Once the fusion protein is obtained in purified form, the protein of interest X is often cleaved from MBP with a specific protease . Protein X can then be separated from MBP by ion exchange chromatography. Orphan date February 2009 Image PBP1.png thumb right 300px Pheromone binding proteins PBPs of insect s are small helix helical protein s postulated to contribute to the specificity of the insect s olfactory system by binding components of the natural pheromone mixtures. Although our understanding of the biochemical mechanisms of chemoreception in insects has greatly improved after the discovery of olfactory receptors and in spite of the significant amount of experimental information available to date for PBPs, the precise role of these protein s is still unknown.
