These tools are used to optimize the design of primers for target DNA or cDNA sequences. Polymerase chain reaction optimization Primer optimization has two goals efficiency and selectivity. Efficiency involves taking into account such factors as GC content, efficiency of binding, complementarity, Nucleic acid secondary structure secondary structure , and DNA melting annealing and melting point Tm . Primer selectivity requires that the primer pairs not fortuitously bind to random sites other than the target of interest, nor should the primer pairs bind to conserved regions of a gene family. If the selectivity is poor, a set of primers will amplify multiple products besides the target of interest ref name BoutrosOkey2004 cite pmid 15073008 noedit ref . This is not the same Pendulum band Pendulum as the drum and bass group that released Hold Your Colour , In Silico Pendulum album In Silico and Immersion album Immersion . Pendulum was a partnership between two Melbourne DJs Noel M and Julian W. They released their first single in 1995 with I Need You Pendulum song I Need You , which reached 1 in Australian Dance charts within three weeks after its release. After its success, Pendulum released Awesome Party Insecurity the following year in 1996. The song was a minor hit compared to I Need You . In 1997, Coma Pendulum song Coma was released proving to be their biggest hit, appearing on the Triple J Hottest 100, 1997 and peaking at 32 on the chart. Extensive support from English and European DJs including Pete Tong and Sasha DJ Sasha gave this song international recognition. Shortly after that release, their only album 3 Knocks was released, which featured all the above singles, new songs, and remixes on two discs. This album was for some time Australia s highest selling Independent release. After the release of 3 Knocks , the group disbanded in early 1998 to pursue other interests. Molecular cytogenetics involves the combination of molecular biology and cytogenetics . In general this involves the use of a series of techniques referred to as fluorescence in situ hybridization , or FISH, in which DNA probes are labeled with different colored fluorescent tags to visualize one or more specific regions of the genome. FISH can either be performed as a direct approach to metaphase chromosomes or interphase nuclei. Alternatively, an indirect approach can be taken in which the entire genome can be assessed for copy number changes using virtual karyotyping. Virtual Karyotype Virtual karyotypes are generated from arrays made of thousands to millions of probes, and computational tools are used to recreate the genome in silico .
