  1. Mycobacterium Tuberculosis Structural Genomics Consortium

Unreferenced date July 2007 The TB Structural Genomics Consortium is a worldwide consortium of scientists developing a foundation for tuberculosis diagnosis and treatment by determining the 3 dimensional structures of protein s from M. tuberculosis . The consortium seeks to solve structures of proteins that are of great interest to the TB biology community. A major goal of the consortium is to have a putative function for every ORF in the TB genome . Activities As of June 2006, 82 TB protein structures have been determined, 15 since January 1, 2006. The database of linked structural genomics structural and functional genomics functional information that has been constructed using this information can form a lasting basis for understanding M. tuberculosis pathogenesis and for structure based drug design. As of June 2006, the TB Structural Genomics Consortium consists of 430 active members in 148 laboratories from 83 institutions across 15 countries. Consortium laboratories are collectively responsible for 3.3 of all protein structures in the Protein Data Bank and have extensive records of methods development. Consortium members have carried out a pilot project on the structural genomics of a hyperthermophile that has identified bottlenecks in the structure determination process and resulted in the development of methodologies for high throughput structure determination and analysis. Nofootnotes article date April 2012 Primarysources article date April 2012 The Structural Genomics Consortium SGC is a not for profit organization formed in 2004 to determine the three dimensional structures of protein s of medical relevance, and place them in the Protein Data Bank without restriction on use. The SGC operates out of the Universities of University of Oxford Oxford and University of Toronto Toronto . Over the past five years, the SGC has accounted for 25 of the global output of novel human protein structures each year, and 40 of the annual global output of structures of proteins from human parasites. The SGC target proteins have relevance to human health and disease, such as diabetes , cancer and infectious diseases such as malaria . The SGC is supported by funding from the Wellcome Trust , GSK , Novartis , Eli Lilly , Pfizer , Takeda , Abbott , Life Technologies , the Canadian Institutes for Health Research CIHR , Canadian Foundation for Innovation CFI , Genome Canada, the Ontario s Ministry of Economic Development and Innovation MEDI . Recently, these organizations together have committed more than US 65 million to the consortium to sustain operation from 2011 2015. The SGC is also spearheading an open access chemistry partnership a new model for pre competitive drug discovery in which the public and private sectors collaborate to generate potent and selective pharmacological inhibitors of human proteins that regulate epigenetic signalling, and commit to make these reagents available without restriction on use. Description Gram positive , nonmotile and acid fast rods. Colony characteristics Variable colony morphology, buff in colour, either rough or smooth. Physiology Growth on glycerol free egg media at 37 C within 28 60 days. May adapt tolerance to glycerol. Sensitive to isoniazid , ethambutol , rifampin , streptomycin and pyrazinamide . Differential characteristics A commercial hybridisation assay AccuProbe to identify members of the M. tuberculosis complex exists. All members of the M. tuberculosis complex share identical 16S rDNA and internal transcribed spacers ITS sequences But molecular differentiation is possible by gyrB sequence polymorphism Pathogenesis Cause of naturally acquired generalized tuberculosis in voles .
