This album was a bit of a departure from the bands previous work. It can be best described as progressive thrash metal , featuring more complex song structures, odd time signatures, numerous key and tempo changes, technical riffs and drumming, and even clean sections that often evoked jazz fusion . This newer, more distinctive style earned the band their biggest critical success to date, and many fans hail it as their finest work. Despite this, the album did not sell as well as their previous works due to a shifting musical landscape and as a result, poor advertisement for the album. The restriction endonuclease PstI recognizes, binds, and cleaves the sequence 5 CTGCAG 3 . However, a recognition sequence refers to a different aspect from that of recognition site . A given recognition sequence can occur one or more times, or not at all on a specific DNA fragment. A recognition site is specified by the position of the site. For example, there are two PstI recognition site in the following DNA sequence fragment, start at base 9 and 31 respectively. A recognition sequence is a specific sequence, usually very short less than 10 bases . An origin of transfer oriT is a short sequence up to 500 Base pair bp of DNA that is necessary for transfer of a bacteria l plasmid from a bacterial host to recipient during bacterial conjugation . The oriT is cis acting it is found on the same plasmid that is being transferred, and is transferred along with the plasmid. The origin of transfer consists of three functionally defined protein domain domains a nicking domain, a transfer domain, and a termination domain. At the beginning of conjugation, a multi protein complex called the relaxosome assembles around the oriT , with each individual protein binding at specific sites on the oriT . A relaxase one of the proteins of the relaxosome nicks the plasmid at the nic site of the oriT, catalyzing a trans esterification reaction that transfers the 5 end of the DNA at the nic site to a tyrosine residue on the relaxase. Relaxase then moves in the 5 to 3 direction on the plasmid, unwinding the DNA in a helicase like fashion, until it comes a full circle back to the oriT where the relaxase recognizes a termination domain that causes it to dissociate from the DNA.
