In analyticalchemistry , samplepreparation refers to the ways in which a sample is treated prior to its analysis. Preparation is a very important step in most analytical techniques, because the techniques are often not responsive to the analyte in its in situ form, or the results are distorted by interfering chemical species species . Samplepreparation may involve solution dissolution , chemical reaction reaction with some chemical species, Powder substance pulverizing , treatment with a complex chemistry chelating agent e.g. EDTA , masking agent masking , Filter chemistry filtering , wikt dilution dilution , sub sampling or many other techniques. The samplepreparation for mass spectrometry is used for the optimisation of a sample for the analysis in a mass spectrometer MS . These have depending on their ion source different requirements for volume, concentration , and composition of the analyte solution. Furthermore, in protein mass spectrometry usually the protein of interest is cleaved into peptides before analysis, either by in gel digestion or by proteolysis in solution. The Society of Public Analysts was formed in the United Kingdom in 1874 and subsequently became the Society for Analytical Chemistry . It was incorporated in 1907. The chemical industry had grown rapidly in the 19th century, and developments in the alkali, explosive and agricultural chemical fields produced a growing need for analytical chemists. Many of these chemists had little or no training in chemistry, and their lack of expertise was a danger to the public. Some time after Parliament of the United Kingdom Parliament passed an Act of Parliament Act to try to remedy the situation, the Society was formed. It published The Analyst chemistry journal The Analyst , Analytical Abstracts and the The Analyst chemistry journal Proceedings of the Society for Analytical Chemistry from 1964 to 1974 . In April 1966 it presented its first Gold Medal to Herbert Newton Wilson in recognition of his contribution to chemical analysis. http pubs.rsc.org en content articlelanding 1966 an an9669100223 unauth It amalgamated with the Chemical Society , the Royal Institute of Chemistry , and the Faraday Society in 1980 to become the Royal Society of Chemistry . One source date December 2010 The slice preparation or brain slice is a laboratory technique in electrophysiology that allows the study of a synapse or neural circuit in isolation from the rest of the brain, in controlled physiological conditions. It involves stimulating and or recording from a slice of brain tissue immersed in artificial cerebrospinal fluid . The technique allows for greater scientific control experimental control , through elimination of the effects of the rest of the brain on the circuit of interest, careful control of the physiological conditions through perfusion of substrates through the incubation fluid, to precise manipulation of neurotransmitter activity through perfusion of agonists and antagonists. However, the increase in control comes with a decrease in the ease with which the results can be applied to the whole neural system. Benefits and limitations When investigating mammalian CNS activity, slice preparation has several advantages and disadvantages when compared to in vivo study. Slice preparation is both faster and cheaper than in vivo preparation, and does not require anaesthesia beyond the initial sacrifice. The removal of the brain tissue from the body removes the mechanical effects of heartbeat and respiration, which allows for extended intracellular recording. The physiological conditions of the sample, such as oxygen and carbon dioxide levels, or pH of the extracellular fluid can be carefully adjusted and maintained. Slice work under a microscope also allows for careful placement of the recording electrode, which would not be possible in the closed in vivo system. Removing the brain tissue means that there is no blood brain barrier , which allows drugs, neurotransmitters or their modulators, or ions to be perfused throughout the neural ... tissue. Policy Debate In policy debate , preparation time prep time is the amount of time given to each team to prepare for their speeches. Prep time may be taken at any time in any interval. Preparation time is also referred to as alternate use time or Batman time. Although preparation time varies from tournament to tournament, in high school each team is generally given between 5 and 8 minutes of prep time depending on the state and tournament in college, each team is generally given 10 minutes of prep time. At some collegiate tournaments, for example the University of Texas at Dallas, alternate use time is used giving the debaters a total of 16 minutes and eliminating the mandatory cross examination periods. This time can be used as preparation time or to ask questions during the normal cross examination periods. Some judges will allow the team taking preparation time to continue asking questions of their opponent. However, because most judges will not require the other team to answer, these questions are generally clarification oriented rather than combative, unlike those asked in cross examination . Many judges disapprove of using alternative use time for non alternate use activities&mdash for example, asking questions of the other team or presenting more arguments.
