A serotonin antagonist is a drug used to inhibit the action at serotonin receptor s. Many of the most important medications of this class selectively act at the 5 HT3 receptor , and thus are known as 5 HT3 antagonist s. However, other drugs such as ketanserin act upon other types of serotonin receptors. Antagonists of the 5 HT2A 5 HT sub 2A sub receptor are sometimes used as atypical antipsychotic s, with the typical antipsychotic s being purely dopamine antagonist s. Allosteric serotonin reuptake inhibitor is a marketing classification invented by the pharmaceutical company Lundbeck for its selective serotonin reuptake inhibitor SSRI escitalopram , the S stereoisomer of the SSRI citalopram . It is based on the observation that the R isomer of citalopram can decrease the potency and inhibit the effects of the S isomer, probably through an allosteric interaction between two distinct, non overlapping binding sites for the two different isomers on the serotonin transporter . The term allosteric refers to the negative interaction between the binding sites of to different drugs or isomers, however, it does not relate to the action of one drug to the inhibition of serotonin uptake. Allosteric serotonin reuptake inhibitors are therefore not a therapeutically distinct class of SSRI. File Serotonin 2D skeletal.svg thumb Chemical structure of the monoamine and indole neurotransmitter serotonin . File Dopamine2.svg thumb Chemical structure of the monoamine and catecholamine neurotransmitter dopamine . A serotonin dopamine reuptake inhibitor SDRI is a drug that acts as a reuptake inhibitor for the monoamine neurotransmitter s serotonin and dopamine by blocking the actions of the serotonin transporter SERT and dopamine transporter DAT , respectively. This in turn leads to increased extracellular concentrations of serotonin and dopamine and, therefore, an increase in serotonergic and dopaminergic neurotransmission . Unlike the case of other combination monoamine reuptake inhibitors such as serotonin norepinephrine reuptake inhibitor s SNRIs and norepinephrine dopamine reuptake inhibitor s NDRIs , on account of the very similar chemical structure s of their substrate biochemistry substrate s, it is extremely difficult to tease apart affinity pharmacology affinity for the DAT from the norepinephrine transporter NET and inhibit the reuptake of dopamine alone. A serotonin receptor agonist is a compound that activates serotonin receptor s, mimicking the effect of the neurotransmitter serotonin . There are various serotonin receptors and ligands. 5 HT sub 1A sub receptor Azapirone s such as buspirone , gepirone , and tandospirone are 5 HT sub 1A sub agonists marketed primarily as anxiolytic s, but also recently as antidepressant s. 5 HT sub 1B sub receptor Triptan s such as sumatriptan , rizatriptan , and naratriptan , are 5 HT sub 1B sub receptor agonists that are used to abort migraine and cluster headache attacks. 5 HT sub 1D sub receptor In addition to being 5 HT sub 1B sub agonists, triptan s are also agonists at the 5 HT sub 1D sub receptor, which contributes to their antimigraine effect. 5 HT sub 1F sub receptor LY 334,370 was a selective 5 HT sub 1F sub agonist that was being developed by Eli Lilly and Company for the treatment of migraine and cluster headaches. Development was halted however due to toxicity detected in animal test subjects. Lasmiditan has successfully completed Phase II clinical trials in early 2010. 5 HT sub 2A sub receptor Psychedelic drug s such as LSD , mescaline , psilocin , Dimethyltryptamine DMT , and 2C B act as 5 HT sub 2A sub agonists. Their action at this receptor is responsible for their hallucinogenic effects. It is now known that many of these drugs act as agonists at many other 5HT receptors in addition to the 5 HT2A including the 5 HT2C and others. 5 HT sub 2C sub receptor Lorcaserin is a thermogenic and anorectic weight loss drug which acts as a selective 5 HT sub 2C sub agonist. 5 HT sub 4 sub receptor Cisapride is a 5 HT sub 4 sub partial receptor agonist that has been used to treat disorders .... A serotonin norepinephrine dopamine reuptake inhibitor SNDRI , or triple reuptake inhibitor TRI , is a drug ligand that concurrently or simultaneously acts as a reuptake inhibitor for the monoamine neurotransmitter s, serotonin 5 HT , norepinephrine noradrenaline, NA and dopamine DA , by blocking the protein action action of the serotonin transporter SERT , norepinephrine transporter NET , and dopamine transporter DAT , respectively. This, in turn, leads to increased extracellular concentration s of these neurotransmitter s and, therefore, an increase in serotonergic , noradrenergic or adrenergic , and dopaminergic neurotransmission . In this sense, to a degree, there is omnipresence in what these compounds do. One might also be of the opinion that they are omnipotent in their mechanism and mode of activity, although it is still debatable to what extent this is true. Unreferenced date December 2009 Serotonergic or serotoninergic means related to the neurotransmitter serotonin . A synapse is serotonergic if it uses serotonin as its neurotransmitter. A substance is serotonergic if it produces its effects via interactions with the serotonin system. A serotonergic , or serotonergic agent , is any chemical which functions to enhance the effects mediated by serotonin in the central nervous system , and they include the following classes of chemicals Serotonin precursors such as tryptophan and 5 Hydroxytryptophan 5 HTP Cofactor biochemistry Cofactor s required in the body s production of serotonin Serotonergic enzyme s Selective serotonin reuptake inhibitor A common class of serotonergic antidepressants Noradrenergic and specific serotonergic antidepressant Another class of serotonergic antidepressants Serotonergic psychedelic s The serotonergic psychedelic drug s See also Serotonin agonist Serotonin antagonist Cholinergic Dopaminergic Nootropic Serotonergics Category Parasympathetic nervous system Category Neurochemistry Category Neurotransmitters Category Serotonin no Serotonerg sr Serotonergik ... more details

In pharmacology , an endogenous agonist for a particular Receptor biochemistry receptor is a compound naturally produced by the body which binds to and agonist activates that receptor. For example, the endogenous agonist for 5 HT receptor serotonin receptors is serotonin , and the endogenous agonist for dopamine receptor s is dopamine . ref Goodman and Gilman s Manual of Pharmacology and Therapeutics. 11th edition, 2008 . p14. ISBN 0071443436 ref In general, receptors for small molecule neurotransmitter s such as serotonin will have only one endogenous agonist, but often have many different receptor subtypes e.g. 13 different serotonin receptor receptors for serotonin . On the other hand, neuropeptide receptors tend to have fewer subtypes, but may have several different endogenous agonists. This allows for a high degree of complexity in the bodies signalling system, with different tissues often showing quite distinct responses to a particular ligand . Some endogenous Antagonist pharmacology antagonists and inverse agonist s are also known e.g., kynurenic acid at the NMDA receptor , but these are much less common.
